CONTRIBUTION TO APRICOT GENETIC ANALYSIS WITH RFLP, RAPD AND AFLP MARKERS

Abstract:
A set of cultivars that included most cultivars used as genitors in apricot breeding programs in Europe were examined by RFLP, RAPD and AFLP molecular marker analyses. A gradation of number of markers from RFLPs (low) to RAPDs (high) to AFLPs (very high) was obtained. For all type of markers the levels of polymorphism obtained were high enough for identification of the cultivars. Correlations were examined between the distance matrices and cultivar groupings based on each marker system. Correlations were higher between RAPDs and AFLPs than between RFLPs and RAPDs or AFLPs. This was due to the number of markers obtained by system and compared. For all DNA-based markers, however, the European cultivars were separated from all or most North American cultivars. AFLP analyses gave the larger number of markers per reaction which allowed the highest resolution for discerning relationships among cultivated apricots.
