PROPAGATION OF VITEX AGNUS-CASTUS BY TISSUE CULTURE

Abstract:
The nutrient media used were WPM and MS supplemented with 0,5 µM IAA and 5 µM ΒΑ. Shoot tip explants were used for establishing in vitro cultures. In the two subsequent subcultures the initial explant was cultured undivided in WPM and MS media again with 0,5 µM IAA and 5 µM BA. In the establishment stage 2-3 new microshoots were formed, while in the 1st and 2nd subculture 5-7 and 11-14 microshoots were produced, respectively. There were no differences between WPM and MS media in terms of microshoot formation in the establishment stage and the 1st subculture, whereas, in the 2nd subculture more microshoots were produced in the MS medium. Microshoots (microcuttings) derived from these subcultures were transferred for rooting to MS medium with 0 or 0,5 µΜ IBA. Rooting was 44% in medium without IBA and 86% in medium containing 0,5 µM of IBA. Rooted microshoots were transplanted to soil mixture in pots and young plantlets were successfully established in the greenhouse.
