Abstract:
Explants taken from axillary shoots -forced or spontaneous- of a 4-year-old selected rootstock of Acer palmatum (cultivar ‘Osakii’) were established in vitro on WPM medium.
Subsequently, new in vitro formed shoots were subcultured and rooted.
Cytokinin TDZ has proved to be useful for establishment and multiplication at a concentration of 0,01mgL-1. Adition of 0,1mgL-1 IAA conjunctly with TDZ had no effect.
Two agar brands were assayed during the multiplication stage (Difco Bacto Agar and Panreac's Agar Bacteriológico Tipo Europeo) and the best results were obtained with DBA. In vitro rooting has been achieved on WPM with adition of 1,0 mgL-1 IBA.
