Abstract:
Several treatments were applied to the apple cv Gala to induce shoot regeneration.
The influence of carbon source, combination and dosage of hormones, kind and age of the explants were evaluated.
The leaves seemed to be the most competent for organogenesis.
Both sorbitol and sucrose induced 50 % regeneration in leaves, while the internode sections were stimulated by the sorbitol.
Glucose had a negative effect on regeneration except for the petioles.
Good results were obtained with leaves in the presence of BAP 22.2 μM and NAA 1.34 μM, with optimal result from apical leaves (about 100 %). Organogenesis was strongly inhibited by TDZ, whereas the combination with BA had an intermediate effect on the explants.
Molecular analysis through RAPDs showed that the regenerants were genetically stable.
