ENDOGENOUS BACTERIA IN TISSUE CULTURES OF CONIFERS - APPEARANCE AND ACTION

Abstract:
Investigations were carried out to find the reasons for difficulties and even declines in different conifer micropropagation systems. Especially in embryogenic lines of Norway spruce and hybrid larch the regeneration capacity was dramatically reduced after repeated subculture periods. A slight clouding of nutrient media gave the first clue for the occurrence of endogenous bacteria. Because of their pleiomorphic growth these bacteria were very difficult to determine and only a crude assignment was possible. The addition of a culture filtrate to embryogenic cultures resulted in a reduced survival of calli. The treatment with the antibiotic cefotaxime could enhance the percentage of clean cultures only for some of the embryogenic lines.
