MICRO-ORGANISMS ACTING IN TISSUE CULTURES OF BLACK LOCUST (ROBINIA PSEUDOACACIA L.)

Abstract:
In the course of developing an appropriate micropropagation method for selected, straight growing genotypes of black locust (Robinia pseudoacacia L.) the enrichment of endogenous bacteria in shoot cultures was observed. It was not possible to suppress the unwanted micro-organisms by adding the antimicrobial stain Malachite Green to the nutrient medium. The bacteria were identified as two strains of Curtobacterium sp. A detrimental effect of the culture filtrate of both strains on callus and shoot cultures was not evident. The survival and growth of micropropagated black locust plantlets during transfer to soil was improved by an early inoculation of rooted shoots in vitro with different nodule forming bacteria strains isolated from older black locust trees.
