Abstract:
The standard procedure for culturing intact ovules on solid medium was compared to two treatments involving the removal of a part of the ovule before culture.
In the latter treatments ovules were cut transsectionally and only the portions containing embryos were cultured.
These portions were either placed with the cut surface in contact with the solid medium, or on solid medium in such a way that the cut surfaces were not in contact with the medium.
These portions were then covered in a drop of semi-liquid medium containing 4 g/L agar.
With all the cultivars investigated, the highest number of embryos was obtained when ovule portions were placed with the cut surface in contact with the solid medium.
