IN VITRO RHIZOGENESIS APTITUDES OF THE PETIOLE OF DIFFERENT GRAPEVINE GENOTYPES - COMPARISON WITH HARDWOOD CUTTINGS

Abstract:
This study was carried out in order to estalish a reliable and reproducible experimental system to obtain roots from the petiole of ten grapevines genotypes in vitro. The petioles with a small blade part (0.5 cm2) were isolated from the mother vitroplant and sub-cultured in Petri dishes. To promote root initiation, four beta-indol butryric acid (IBA) concentrations (0, 0.5, 1, 1.5 mg 1 -1) were tested. In a second experiment, two sucrose contents (10 and 20 g 1-1) were combined with two IBA concentrations (0 and 1 mg 1-1). The petiole of most of the genotypes tested presented rhizogenesis aptitudes. The firts roots appeared 10 days after sub-culture. The average root fresh wieght was 20 mg per dish 15 days later.The addition of IBA increased the root number and/or the root total fresh weight of root depending on the genotype. When sucrose content of the medium was lower (10 mg 1-1), the rooted petiole number increased. Cabernet Sauvignon, Fercal, 101–14. 110R and Rupestris du Lot initiated roots very quickly and had the most homogeneous response whereas 140Ru, Riparia Glorie and 333EM did not root so easily, and SO4 presented very poor aptitudes under these conditions. There was no significant correlation between the rhizogenesis aptitudes of petioles in vitro and of hardwood cuttings, except for the fresh weight per root. The in vitro methodology described in this paper made it made it possible to obtain rapidly a developing root system for most of the paper made it possible to obtain rapidly a developing root system for most of the genotypes studied.The suitability of this experimental system to study the physiological characteristics of the root under variuos growth conditions or the host-parasite relationships has now to be tested.
