ISHS Acta Horticulturae 528: VII International Symposium on Grapevine Genetics and Breeding

EFFECT OF HYGROMYCIN, KANAMYCIN AND PHOSPHINOTHRICIN ON THE EMBRYOGENIC CALLUS DEVELOPMENT AND AXILLARY MICROPROPAGATION OF VITIS VINIFERA L.

Keywords:   Vitis vinifera, selective agents, embryogenesis, micropropagation, genetic transformation
Abstract:
The effect of three selective agents, hygromycin, kanamycin and phosphinothricin, on the embryogenic cultures growth of four cultivars of Vitis vinifera was studied. Only the first two could significantly reduce the growth without inducing whole necrosis of the embryogenic calli. In complementary experiments, the response of axillary buds to hygromycin and kanamycin was evaluated for eight cultivars of Vitis vinifera. As for the embryogenic callus, hygromycin appeared more phytotoxic to axillary buds than kanamycin, but the toxic effects of both antibiotics were different.
