VARIETAL IDENTIFICATION USING PROCEDURES OF STEPWISE DISCRIMINATION

Abstract:
As an alternative to the recognized and validated methods of DNA analysis adopted for the identification of vines, a low cost method based on the acquisition of simple parameters is proposed. The analysis followed a stepwise procedure, with the successive introduction of single characters as discriminatory elements. Characters of a different nature were considered (biochemical markers, morphological descriptors), chosen on the basis of criteria of simplicity, confidence and repeatability of the determination: when possible the expressions of the character were ascribed to only two clearly distinguishable alternatives (e.g. presence/absence). As to the order in which a character was placed in the sequence of levels of choice, decisions were taken on the basis of the relative discriminatory efficiency ascertained in previous experiments. An overall appreciation of the method was therefore obtained a posteriori, comparing the results of different combinations of characters. Maximum efficiency was reached differentiating the varieties with respect to the isoenzymatic forms of GPI and PGM, berry colour and thereafter a few ampelographic characters of the bud, leaves, etc. For the final step, a second procedure was tested to evaluate the discriminatory capacity of some ampelometric parameters of the leaf. The two procedures were compared and a judgement given in terms of confidence of the method.
