INVESTIGATIONS OF GRAPE ANTHER CULTURE AIMING HAPLOID PLANT PRODUCTION

Authors:   R. Holló, S. Misik
Keywords:   Vitis, microspore, microscopy
Abstract:
The effect of the developmental stage of the explants, pretreatment of the inflorescences and medium composition on the embryogenesis and green plant regeneration in grape anther culture was investigated. Three varieties were observed: Mädchentrauben, Rhein Riesling and Rupestris du Lot. Mädchentrauben proved to be recalcitrant in all cases. Rupestris du Lot showed higher embryogenesis than Rheinriesling. Haploid regeneration was not observed. Intensity of somatic embryogenesis of Rupestris du Lot was the highest at sporogen-PMC stages on 2% sucrose with 10-day coldtreatment for both field and forced material. Intensity of somatic embryogenesis of Rheinriesling was the highest at sporogen-tetrad stages for field material and at mid-late uninucleate stages for forced material.
