Abstract:
The isolation of high molecular weight DNA (HM-DNA) is an important step for the construction of physical maps by pulsed field gel electrophoresis and for preparing yeast or bacterial artificial chromosome libraries.
This report describes a new method for preparing high molecular weight DNA from protoplasts and resolution by pulsed field gel electrophoresis in Vitis vinifera.
A high yield of HM-DNA useful for digestion with restriction enzymes, separation by pulsed field gel electrophoresis and hybridization with digoxigenin labeled probes is obtained.
For hybridization specific molecular probes from known Vitis genes have been developed.
