Abstract:
A progeny was obtained from a cross between Moscato bianco and Vitis riparia. Plants were multiplied by cuttings in order to best evaluate the expression of quantitative characteristics and among them fungus-resistance or tolerance.
In a first approach, to test polymorphism and segregation of molecular traits in the progeny, microsatellite and RAPD markers were generated from the DNA of parental lines and 90 seedlings.
Simple sequence repeats alleles were fluorescently tagged by polymerase chain reaction amplification with dye-labeled primers and run by capillary electrophoresis on an automated DNA analyser using fragment-sizing software.
Parental allele sizes were different at all loci analyzed.
Among the offspring, 8 seedlings were the result of the Moscato bianco self-pollination while the rest were hybrids.
The allele segregation ratios were close to the expected Mendelian values and all loci except VVMD6 and VVMD7 turned out to be independently inherited.
RAPD markers segregation in the progeny was less useful for mapping purposes because with time, difficulties were encountered in the reproduction of the banding patterns.
