POTENTIAL ROLE OF OXIDATIVE STRESS IN THE RECALCITRANCE OF GRAPEVINE PROTOPLASTS

Abstract:
Protoplasts from most of the important agricultural plant species, including grapevine, exhibit recalcitrance to plant regeneration. The possible implication of oxidative stress in the recalcitrance of plant protoplasts was studied, using the recalcitrant grapevine protoplasts and as a reference model, the regenerating and non-regenerating tobacco protoplasts. The quality of the commercial cell wall hydrolysing enzymes, used for protoplast isolation, was tested on active oxygen species (AOS) production. Contaminants of cell wall hydrolysing enzymes, such as xylanase, acted as elicitors for production of active oxygen species during protoplast isolation. The enzymic scavenging activity in regenerating and non-regenerating protoplasts was compared. Total peroxidase, NADH peroxidase, catalase, superoxide dismutase, ascorbate peroxidase and glutathione reductase activities and/or isoenzyme profiles were studied during the isolation and the culture of protoplasts. Significant differences were found in the enzymic activities and the expression of specific isoforms between the regenerating and non-regenerating protoplasts.
