GRAPEVINE (VITIS VINIFERA L.) ALCOHOL DEHYDROGENASE GENE: AN IMPORTANT MARKER OF THE PHYSIOLOGICAL STATE DURING FRUIT RIPENING

Abstract:
At the inception of grape berry ripening, when sugar import increases and whereas respiration rate is maximal, a strong increase of ADH activity and of its RNA trancripts occurs. Thus, grape berry Adh expression is developmentally-related. To get insight the molecular mechanisms involved, we focused on the cloning of the Adh cDNAs expressed during ripening. Studies of Adh expression during fruit development and in different organs is presented. Adh transcripts are principally detected in ripening fruits, whereas constitutive or no expression was found in young expanding leaves, tendrils and roots. One grapevine (Vitis vinifera L.) Adh gene, related to fruit ripening and referred to here as GV-Adh1, has been completely sequenced. We report here the structural characteristics of this gene. The 1140 nucleotides open reading frame of GV-Adh1 codes for a predicted 380 amino acid polypeptide with 41 kDa molecular mass and 6.9 isoelectric point. GV-Adh1 deduced protein is closer to those of apple tree and strawberry than to other dicots. Phylogenetic analysis suggests that Vitaceae and Rosaceae could share a common ancestor from the Adh evolution point of view.
