MUTATION INDUCTION BY ION BEAM IN ORNAMENTAL PLANTS - EFFECTS ON SEED GERMINATION AND DNA POLYMORPHISM IN EUSTOMA GRANDIFLORUM

Authors:   S. Ohki, Y. Sakai, K. Chatani, K. Koreeda
Keywords:   Eustoma grandiflorum, DNA polymorphism, ion beam, mutation breeding, RAPD
Abstract:
Seeds of Eustoma grandiflorum were irradiated under aseptic conditions with a 50-MeV 4He2+ beam 3 and 10 days after imbibition. The seeds that were irradiated 3 days after imbibition were not affected by the irradiation. However, less than 5% of the seeds irradiated 10 days after imbibition survived after 4 weeks in culture. At the time of irradiation, these seeds already had elongated radicles. Total DNA was analyzed using 10-mer random primers. DNA polymorphism was detected in the plants from ion beam-irradiated seeds.
