CYCLAMEN PERSICUM MILL. SOMATIC EMBRYOS DEVELOPED IN SUSPENSION CULTURES: HISTOLOGICAL ANALYSIS AND CONVERSION TO PLANT

Abstract:
To develop a fast and easy reproducible propagation system for Cyclamen persicum Mill. hybrids via embryogenic suspension cultures, maturation, germination and conversion to plant have been studied. The presence of abscisic acid (10 μM) in the maturation medium did inhibit secondary embryogenesis. A combination of 10 g/l of mannitol and 10 mg/l of glutamic acid increased the germination percentage up to 61. A histological and histochemical analysis of the structures during differentiation was performed. Accumulation of starch, lipids and phenols inside the cells of the tuber and the formation of vascular elements were demonstrated.
