Abstract:
Matriconditioning with Micro-Cel E as the newest technique of presowing seed treatment was applied in order to improve seed germination.
Cytological and physiological changes during seed ageing followed by matriconditioning were examined to explain repairing activity of the treatment.
Cucumber seeds were aged at 42°C and 96% RH to lower the germination down to 90, 70, 50, 30% and then were matriconditioned with Micro Cel E (MC) for 6 days.
Germination rate and ability, soil emergence, cytological investigation in light microscopy and physiological changes as dehydrogenase activity, in vivo ACC oxidase actitivity, endogenic ethylene production and electrolyte leakage were performed.
Matriconditioning improved germination rate of cucumber seeds by stimulation of hydrolysis of storage protein bodies in embryo, increasing dehydrogenase activity, endogenic ethylene production, ACC oxidase activity and decreasing electrolyte leakage of seeds.
Seeds of the lowest germination percentage showed a decrease of enzyme activity and increase of electrolyte leakage connected with damage and necrosis of cells, growth of pathogenic fungus and necrotisation of whole seeds.
The results indicate that matriconditioning is a successfull method to improve the quality of aged seeds if the deterioration of seeds had not gone too far.
