Abstract:
Well-developed first or second true leaves, originating from in vitro plants of two Cucumis species (C. melo, C. metuliferus) and three Cucurbita species (C. ficifolia, C. maxima, C. pepo), were used for protoplast isolation.
The optimal composition of the enzyme solution and the most suitable isolation method were studied.
The density and viability of protoplasts were used as efficiency criteria.
The best isolation method and the enzyme solution were more suitable for Cucumis spp. than for Cucurbita spp., as the former had more viable and less damaged (plasmolyzed) protoplasts.
