AGROBACTERIUM-MEDIATED TRANSFORMATION IN CUCUMIS SATIVUS VIA DIRECT ORGANOGENESIS

Abstract:
Transformation of cucumber cv. Pointsett 76 was attempted using Agrobacterium tumefaciens via direct organogenesis from cotyledons. Low concentrations of BAP produced shoots and roots from the proximal ends of the cotyledons. Cucumber cotyledon explants were cocultivated with disarmed Agrobacterium strain EHA 105 pME 524(pGA492 Gl - Bar(-). The T-DNA region of this vector contains neomycin phosphotransferase II (npt-II), beta-glucuronidase (uidA) and bar genes. Transformed shoots were more efficiently selected by 6mg/1 PPT. Shoot regeneration occurred within 4 weeks. Acetosyringone enhanced the efficiency of transformation of cotyledon explants.
