DIVERSITY OF NUCLEAR DNA CONTENTS OF HYDRANGEA

Abstract:
Information about Hydrangea ploidy and DNA content is poor. Species are described as diploid 2x = 36 (Sax 1931, Sugiura 1931). This study was carried out to characterise Hydrangea species and cultivars from the French collection settled in Angers.

Flow cytometry was used with a monoparametric cytometer, using DAPI (6-diamino-2phenylindole, A-T specific fluorochrome). Fluorescence of Hydrangea nuclei was compared with an internal reference of Pisum nuclei. Direct determination of chromosome number from root tips with a microscope made possible the calibration of the flow cytometer.
