STRUCTURAL AND MICROANALYTICAL STUDIES ON FROZEN-HYDRATED TISSUES FROM DIFFERENT KIWIFRUIT ORGANS

Abstract:
The analysis of ion distribution and accumulation at various time of plant development and for different physiological conditions in fruit tree organs is fundamental to understand the quali-quantitative mineral requirement in order to enhance fruit production. Biochemical assays cannot easily provide the spatial localisation of mineral elements distribution and storage at cell-tissue level. To overcome these problems and provide a reliable pattern of ion content and distribution, cryofixed samples maintained in their frozen-hydrated state can be analysed by Low Temperature Scanning Electron Microscopy (LTSEM) and EDX X-ray.

In this article, some preliminary structural and analytical investigations performed with LTSEM and EDX X-ray microanalysis on frozen-hydrated kiwifruit leaf, peduncle and fruit samples will be presented.
