FIRST STEP IN THE SEARCH FOR AFLP MARKERS LINKED TO SEX IN ACTINIDIA

Abstract:
Bulked segregant analysis and AFLP (Amplified Fragment Length Polymorphism) technique were used to search for molecular markers linked to sex in Actinidia. The DNA bulk pairs consisted of male and female progeny respectively of either an open-pollinated diploid A. chinensis genotype or an interspecific cross A. deliciosa x A. eriantha. AFLP markers were produced with 64 EcoRI/MseI primer combinations. More than 4000 AFLP loci and more than 100 polymorphic bands were observed. The number of AFLP loci varied with population, bulk and primer combination used, ranging from 7 to 100 for A. chinensis and from 50 to 130 for the A. deliciosa x A. eriantha cross, while the number of polymorphisms between male and female bulks ranged from 1 to 13 for both families. Well-separated and polymorphic AFLP bands were obtained in 12 of the 64 primer combinations assayed. For these primer pairs, the analysis was extended to each single individual of the bulks. A total of four bands have been found apparently linked to sex: two bands were found exclusively in male and two mainly or exclusively in female individuals. The verification of these putative sex-linked AFLP markers on a large sibling progeny as well as the conversion of the sex-linked AFLP markers into locus-specific SCAR markers is both in progress.
