PRODUCTION OF PROBES FOR THE CHESTNUT GENOTYPES BY RAPD MARKERS

Abstract:
The rapid generation of probes for several chestnut cultivars by the RAPD technique is a method that does not require any preliminary information on the genome of the species to be investigated. The procedure is based on the production of RAPD fingerprinting. The amplification product(s) peculiar to one cultivar or common to two or more types of chestnut can be purified and used as molecular probes in hybridisation experiments for the identification of cultivars.

We tested this methodology on chestnut genotypes by amplifying the total DNA extracted from several chestnut cultivars.
