PECTIC STATUS AND CALCIUM NUTRITION IN MICROPROPAGATED CHESTNUT SHOOTS

Abstract:
Micropropagated shoots represent a convenient model for the study of lignified plants. This system reduces their culture cycle to only 3 or 4 weeks thus rapidly producing material for experiments. We use this model in order to better understand the relation between pectic metabolism and calcium variations in the culture medium. The results presented here concern the structure of pectins produced by chestnut shoots grown under different calcium concentrations. First, no real difference is observed in extraction yield. Next, polymersare is separated by ion-exchange chromatography. Each time, at least two fractions are separated: one is neutral and one or more are acid. The neutral fractions contain lower uronic acid percentages (from 0 to 17 %) than the others. On the other hand, galacturonic acid represents at least 40 % of the total sugars in acid fractions. The problem now is to find out if the acidity of pectins can be correlated with the variations of calcium content measured in the tissues in relation with supplies.
