EFFECT OF ROOT LENGTH AND ROOT MACERATION TIME ON BANANAS (MUSA AAA) PLANT-PARASITIC NEMATODE RECOVERY

Abstract:
Three experiments were conducted to evaluate the effect of root length and its maceration time on R. similis, Helicotylenchus spp., Meloidogyne spp. and Pratylenchus spp. recoveries utilizing the root maceration method. The first experiment included four root lengths (1, 2, 3 and 4 cm) and three maceration times (10, 20 and 30 sec) in a completely randomized design, with 10 replicates. The second evaluated the same root lengths, macerated for 10 sec in a completely randomized design with 30 replicates. In the third, five root lengths from 1 to 5 cm were evaluated, using a maceration time of 10 sec in a Latin square design with 30 replicates. Nematodes were extracted from 25 g root samples and expressed by 100 g of roots. The results indicated that root lengths and root maceration times did not affect (P > 0.05) nematode recovery. Therefore, it is suggested to use a root length from 1 to 3 cm, and a maceration time of 10 sec in low and 10 sec in high speed. This recommendation will allow to increase the laboratory processing speed and to reduce the labor accident risks, holding up the nematode recovery efficiency.
