MICROPROPAGATION OF GERBERAS FROM FLORAL BUDS

Abstract:
We have obtained more than 1000 Gerbera hybrids in Colombia. Some of them show excellent agronomic characters such as color, floral diameter, stem length and vigor. As the classical propagation technique through lateral shoots is not very efficient, we studied the factors affecting the in vitro regeneration from floral buds, multiple shoots micropropagation and rooting. Young floral buds of 1 cm or less were surface sterilized and incubated on a basal MSH medium containing half strength Murashige & Skoog (MS) macroelements salts, NaFeEDTA 25 ppm, Heller's (H) microelements, vitamins, 10 ppm BAP (Bencylaminopurine) and 0,1 ppm IAA (Indoleacetic acid). Four weeks later the explants were transferred on a modified medium with a reduced concentration of BAP (2mg/L). Four weeks later, the regenerated shoots were finally transferred separately to a medium without hormones in order to induce the plantlet regeneration. We evaluated four concentrations of BAP and a control (0, 1, 2, 4 and 8 ppm) during the micropropagation phase. The best results were obtained with 1 or 2 ppm BAP. Higher concentrations (4 and 8 ppm) reduced noticeably the shoot proliferation. For the rooting induction, we evaluated two salt concentrations of MSH (normal and ½ strength) in combination with 0 and 1 ppm IAA on a randomized design. The best treatment for the rooting induction and plantlet development was the one with half the strength of salts without IAA.
