Abstract:
An efficient micropropagation system using disinfectant incorporated medium was developed.
In the production of in vitro plants, much labor and time are necessary for preparing the medium and for transplanting the somatic embryos into it under aseptic condition.
With the new culture method, sodium hypochlorite (active chlorine 5–10 ppm) and streptomycin (25 mg/l) were incorporated into the plant regeneration media (½ MS + sucrose 2%) and embryos were transplanted in open air condition.
The concentration of these disinfectants did not damage the plantlets and suppress the contamination.
This method was also available for liquid germination media of somatic embryos and for that of plant regeneration using novel fluorocarbon polymer film culture vessel (PCP).
