DETECTION OF EXTRACELLULAR PROTEINS SECRETED BY ASPARAGUS EMBRYOGENIC CELLS IN SUSPENSION CULTURES

Abstract:
The effect of plant growth regulator (PGR) concentration and length of time in culture, on the expression of extracellular proteins secreted into the medium of asparagus (Asparagus officinalis L.) embryogenic cell cultures was investigated. Embryogenic calli were initiated on Murashige and Skoog (MS) medium containing 0.3, 1.0, 3.0 and 10μM 2,4-D plus 1.0μM NAA/0.1μM kinetin. Embryogenic cell suspensions were obtained by transferring embryogenic calli into PGR-free liquid medium. Extracellular proteins were extracted from media of actively growing embryogenic suspension cultures and analyzed by gel electrophoresis. Extracellular proteins between 14 and 97 kD were detected in the culture medium of cultivars 'Aspiring', 'Karapiro', 'Turoa' and 'Pacifica' and could be used to distinguish each asparagus cultivar investigated. The PGR concentration used to initiate embryogenic calli and the length of time cells were maintained on PGR-free medium were shown to influence the expression of extracellular proteins. Further studies are required to determine the effect of these extracellular proteins on somatic embryo development.
