Abstract:
The aim of the study was to determine the influence of lowered nitrogen concentration and 2iP vs.
BA on plum (Prunus domestica L.) shoot proliferation and elongation.
Full-strength solid MS-medium, containing 20,6 mM 1-1 NH4+ and 39,4 mM 1-1 NO3- was compared with MS-medium, supplied with 10,3 mM 1-1 NH4+ and 29,1 mM 1-1 NO3- In both cases either 4,44 μM 1 -1 BA or 49,21 μM 1-1 2iP was added into medium.
Lowered nitrogen concentration did not affect the proliferation efficiency and shoot elongation.
There was a difference in plantlets appearance in vitro - shoots on full-strength MS medium had dark-green leaves while in lower nitrogen concentration light-green leaves developed.
BA induced shoot proliferation, not depending on the concentration of nitrogen in culture medium. 2iP strongly inhibited formation of adventitious shoots but had an elongating effect.
Shoots treated with 2iP were more ligneous and acclimatised more successfully when transferred to greenhouse for rooting in vivo.
