Abstract:
A radioimmunoassay protocol for abscisic acid (ABA) determination in olive (Olea europaea L.) leaves (cv Frantoio), based on a fast and one-step purification of the extract, is described.
DBPA1, an ABA highly-specific monoclonal antibody, was used.
Extract purification was obtained by means of subsequent elutions with H2O, 5% and 100% methanol in 1-ml C18 silica prepacked cartridges.
This procedure made it possible to eliminate competitive interferences (approximately 20%) with DBPA1 for ABA, as revealed by HPLC fractioning of crude aqueous extracts.
Experiments with tritiated ABA indicated that the use of an 8-sample cartridge rack ensured high percentages of ABA recovery, as well as good experimental reproducibility.
The efficiency of the purification procedure was confirmed by comparing ABA levels in olive leaf extracts, purified by C-18 cartridges or by HPLC.
