A LINKAGE GENOME MAP FOR OLIVE AS AN IMPORTANT TOOL FOR MARKER-ASSISTED SELECTION

Abstract:
A genomic linkage map represents an important tool to use in olive breeding and to select new genotypes obtained by cross or by gene transfer. To construct the map two important steps have to be considered: the availability of numerous and reproducible molecular markers and populations segregating for important agronomic traits. As marker systems both RFLP (Restriction Fragment Lenght Polymorphism) and AFLP (Amplified Fragment Length Polymorphism) were considered: RFLP as codominant markers and AFLP as fast and easy to handle markers. As segregating population an F1 progeny was used, because the long juvenile phase of olive makes it difficult to obtain backcrosses or F2 progenies. There are some restrictions in using F1 populations for map construction because only the markers heterozygous in one parent and homozygous recessive in the other or heterozygous in both parents can be utilized. Moreover, F1 progenies generate one map for every parental line and only double heterozygous markers make it possible to join information derived from different maps. Some crosses, segregating for different characters, were considered. We report our first experience on the cross Leccino x Dolce Agogia, segregating for the Stearoyl-ACP Desaturase gene. 100 individuals of this F1 population were screened using both AFLP and RFLP markers. AFLP showed a high level of polymorphism. In each gel about 20 heterozygous bands per lane were obtained, 85% of which were useful to generate marker loci to locate on the map. The linkage groups, obtained with the MAPMAKER program, are shown.
