MERISTEM TIP CULTURE AND HEAT THERAPY FOR PRODUCTION OF APPLE MOSAIC VIRUS FREE PLANTS IN INDIA

Abstract:
Apple meristems ranging in size from 0.1 to 1.0 mm were cultured on medium supplemented with BA 1.0mg/L, IBA 0.1mg/L and GA3 0.1mg/L. As the meristem size increased, the rate of meristem survival and shoot regeneration increased. Apple mosaic virus, however, could be detected in cultures raised from meristems longer than 0.2 mm. Complete virus inhibition was also observed with hot water treatment of wood scions at 47°C for 30 minutes and 50°C for 15 minutes. Hot air treatment of potted plants at 37°C for 4 weeks and 40°C for 2 weeks also resulted in complete virus elimination.
