ISOLATION AND CHARACTERIZATION OF CDNAS DIFFERENTIALLY EXPRESSED DURING EARLY PEACH [PRUNUS PERSICA (L.) BATSCH] FRUIT DEVELOPMENT

Abstract:
We are interested in determining genes which are potentially involved in the control of peach [Prunus persica (L.) Batsch] fruit development and acidity.
Two different approaches were used.
First, we looked for genes already known to be involved in the synthesis of organic acids.
Using a RT-PCR strategy, we isolated a partial cDNA corresponding to phosphoenolpyruvate carboxylase (PEPC). In a second approach, cDNAs for genes differentially expressed during the first growth phase of the fruit, when malic acid strongly accumulates, were isolated from a library made from young peach (cv.
Fantasia) fruit, 32 days after bloom.
A total of 32 clones were obtained and sequenced.
The cDNAs presenting homologies with known sequences range in two groups: defense-related genes (metallothionein, protease inhibitor) and development-related genes (ADP-ribosylation factor, translationaly-controlled tumor protein, ribosomal proteins). The expression patterns of these genes have been investigated in young leaf, flower and throughout peach fruit development.
Most of the clones studied are highly expressed in fruit, during the first phase of peach fruit development.
