CRYOPRESERVATION OF VEGETATIVELY PROPAGATED TROPICAL CROPS BY VITRIFICATION

Abstract:
In the tropics, there are many important crops propagated vegetatively. Most of the germplasm of these species is mainly conserved in field gene banks and there is a growing need for stable long-term storage of the germplasm. Although recent remarkable progress in cryobiological studies of plant materials has made cryopreservation a realistic tool for long-term storage, cryopreservation of tropical species has been less extensively investigated when compared to temperate species. In this regard, more studies are required for tropical species. In this report, cyostorage procedures of shoot tips by vitrification were investigated and compared for different species such as taro, banana and yams. Optimized protocols were applied to different genotypes and an average post-thaw survival of 80, 70 and 63% was obtained for taro, banana and white yam, respectively. The studies demonstrated that cryopreservation of shoot tips by vitrification can be applied to tropical crops.
