DEVELOPMENT OF PILOT SCALE PROCESS FOR MASS PRODUCTION OF LILIUM BULBLETS IN VITRO

Abstract:
Using a two-stage culture process separating bulblet formation and the development stage, the possibilities for a mass propagation system by bioreactor were investigated. The nutrient requirements for bulblet formation and development were varied for each stage. It was revealed that each stage could be controlled by the manipulation of nutrients and culture conditions. By using a two stage culture process, various types of bioreactors were employed for mass production of bulblets. Several thousand bulblets were produced in 5–20 liter batches of non-stirred bioreactors within 60 days. In addition, a considerable increase of bulblet weight was observed. With the passage of time, sugar and nitrogen were consumed rapidly. In particular ammonium was used more preferentially as a nitrogen source than the nitrate ion for bulblet growth.
