MOLECULAR ANALYSIS OF BRASSICA CMS AND ITS APPLICATION TO HYBRID SEED PRODUCTION

Abstract:
The oilseed rape species Brassica napus contains two endogenous male sterile cytoplasms, nap and pol. Studies of mtDNA organization and expression have led to the identification of mitochondrial gene regions thate seem likely to specify these forms of male sterility. Each of these regions contains a novel gene, and expression of each region is affected by the corresponding nuclear restorer gene, Rfn for nap CMS and Rfp for pol CMS. To assess the capacity of orf224, the novel gene associated with pol CMS, to induce male sterility, we fused the gene to a sequence encoding a mitochondrial transit peptide, and introduced this sequence into the nuclear genome of male fertile B. napus plants. Plants with markedly reduced male sterility were recovered. These observations suggest a strategy for addressing some of the practical problems encountered with hybrid seed production using these CMS systems.
