Abstract:
The first programme of the molecular biology laboratory of BBV was concerned with the construction of an RFLP map of the Brassica oleracea genome (cauliflower and broccoli). The current phase of work consists of locating interesting genes in this map.
We report here on the mapping of a nuclear recessive gene which encodes for a male sterility trait.
We have sought to obtain molecular markers closely linked to that gene, with a view to applying Marker Assisted Selection (MAS) to cauliflower and broccoli breeding programmes.
RAPD markers were preferred for their ease of use.
Four markers were obtained by applying Bulked Segregant Analysis to 5 segregating populations (4 F2s and 1 DH ; surprisingly, we found that the male sterility gene was dominant in one of the 4 F2s, although its parental F1 hybrid plant was male fertile). Two markers have been located in the RFLP map.
The use of the markers in MAS is underway.
