IN VITRO MYCORRHIZAL SYNTHESIS OF MICROPROPAGATED TILIA PLATYPHYLLOS SCOP. PLANTLETS WITH TUBER BORCHII VITTAD. MYCELIUM IN PURE CULTURE

Abstract:
A method for in vitro ectomycorrhizal synthesis using micropropagated Tilia platyphyllos Scop. plantlets and Tuber borchii Vittad. mycelium in pure culture was developed for use as a model system in future biotechnological research. The Tilia micropropagation was obtained from buds of a single plant in order to limit somaclonal variations and T. borchii mycelium was isolated from a single fruit-body. The T. borchii mycelium was inoculated into test tubes, previously filled with a mixture of peat-moss and vermiculite embedded with MS/2 solution, containing a rooted linden seedling. Completely formed ectomycorrhizae were observed after three months. The anatomical and morphological features of the ectomycorrhizae observed using light and confocal microscopes were identical to those usually obtained under non-sterile conditions. The mean degree of ectomycorrhizal formation was high (86.64 ± 10.09 %). ITS-PCR analyses confirmed that the ectomycorrhizae obtained were of T. borchii.
