Abstract:
We have developed an efficient technique for shoot regeneration of lisianthus from leaf explants by direct organogenesis.
Transformation experiments with DNA were carried out using Agrobacterium tumefaciens carrying the Ti plasmid pBI121 which contained the genes NPTII and ( -glucuronidase) GUS fused to the 35S promoter of the CaMV. Kanamycin-resistant shoots of lisianthus were regenerated on Murashige and Skoog (MS) medium with 6-Benzylaminopurine (BA) showed GUS activity, as determined by the appearance of a blue precipitate after incubation with the substrate X- Gluc (5-bromo-4-chloro-3-indolyl glucuronide).
