TISSUE CULTURE METHODS AND CLONING OF THE NEEM TREE (AZADIRACHTA INDICA) FOR BIOINSECTICIDE PRODUCTION

Abstract:
Hypocotyl explants of the Neem tree, Azadirachta indica A. Juss (Venezuelan variety) developed callus and shoots when cultured on Murashige and Skoog (MS) medium supplemented with IAA and BA. Axilary buds and shoot explants developed a light yellow friable callus when incubated in the dark at 25±1°C. Shoots were regenerated from nodal buds and calli. About 60% of the shoots developed roots when transferred to MS medium without BA. Rooted plants transferred to the greenhouse, acclimated and developed into sizable plants. The best culture medium for root development in shoot explants was MS medium without BA but with 5 μM of IAA. Somatic embryogenesis from callus cultures was observed. Bioassay tests were performed to evaluate the effect of leaves, callus and liquid grown Neem tissue and medium extracts on the feeding behavior of desert locust larvae. Bioassay tests indicated that in vitro plants can be a source of antifeedant compounds of the Neem tree.
