MICROPROPAGATION OF SELECTED PETASITES HYBRIDUS (L.) PLANTS WITH HIGH PETASIN CONTENT IN LEAVES

Abstract:
22 populations were screened for plants with high petasin content. Rhizomes and leaves were separately analyzed. Two populations revealed plants with more than 10 mg / g Dw petasin in leaf matter. Leaves are almost void of toxic PA in contrast to the underground organs. Floret buds from these selected clones were collected in march and micropropagated on a solid MS medium performing a two step protocol. Shoot induction occured with 17.6 μM BA + 0.54 μM NAA. Arising shoots were excised after 5 weeks of culture and subcultured on a shoot multiplication medium with 8.8 μM kinetin + 0.54 μM NAA. After 7 – 8 weeks shoots were transferred directly to soil or rooted on ½ strength MS medium prior to ex vitro culture.
