Abstract:
Embryogenic callus, normal plantlets and abnormal plantlets could be induced from immature embryos cultured on MS basal medium for one month.
The growth of embryogenic callus on the MS basal medium was better than B5 and White medium.
When embryogenic callus was cultured in liquid MS medium, it formed suspension cells and the cell clumps were keeping on growing and developing further more into somatic embryos and seedlings.
Adding 0.3% agar or 0.1% gelrite to the liquid medium would help to form more somatic embryos and seedlings.
When the seedlings were cultured on filter paper rinsing by liquid ½ MS medium contained 3% sucrose, the surviving rate was around 40% in vitro and 30% after transplanting.
Plantlets originated from in vitro culture showed morphological similarities with original plants.
Furthermore, TLC of the plantlets originated from in vitro culture was similar to that of A. sinensis sold in market.
