EFFECT OF CA2+ RELATED WITH NAA AND BA ON ADVENTITIOUS BUD FORMATION IN MIDVEIN SEGMENTS OF HYBRID POPULUS (POPULUS NIGRA VAR. BETULIFOLIA X POPULUS TRICHOCARPA).

Abstract:
Midvein segments of poplar were cultured in WPM with 0.2 mg/l of BA and 0.01 mg/l of NAA and were transferred WPM with NAA and without BA or WPM with BA and without NAA in every 3 days adventitious bud formation after that BA was activated.

To know the relation of calcium and hormone action in adventitious bud formation, 0.1 μM of A23187 or, 1mM of EGTA or, 100 μM of W-7 was added to each WPM with 0.01 mg/l of NAA and 0.2 mg/l of BA, WPM with 0.01mg/l of NAA and without BA, and WPM with 0.2mg/l of BA and without NAA. Adventitious buds formation was increased by A23187, calcium synergist, and decreased by EGTA, calcium antagonist. The effects of 0.1 μM of A23187 and 1mM of EGTA on the adventitious bud formation was increased in segments cultured in WPM with 0.2 mg/l of BA.

A calcium synergist A23187 supplementation of WPM, especially with 0.2 mg/l of BA, further improved the regeneration potential of midvein explants.

Overall, the results suggest that calcium should be highly correlated with BA and NAA in adventitious bud formation in midvein segments of hybrid Populus.
