Abstract:
Quercus faginea LAMK. acorns were collected three times every 10 days during the summer.
Zygotic embryos were cultured in darkness in a MS medium replaced by PRL-4-C macrosalts and adding different concentrations of NAA, BA and 2,4-D. 30 days later, explants were transferred to basal medium containing 0,5 μM BA and 0,5 μM NAA in light. 30 days later, explant were transferred to a PRL-4-C medium.
Somatic embryogenesis appeared on the explants at the end of the third month in culture.
Earlier acorn maturation stages gave higher rates of somatic embryogenesis than later stages.
Somatic embryogenesis appeared directly on the zygotic embryos without detectable callus.
Secondary embryogenesis appeared subculturing these somatic embryos during 8 months.
