PLANT REGENERATION FROM CAMBIUM EXPLANTS OF THE HYBRID ELM 'SAPPORO GOLD 2' (U. PUMILA X U. JAPONICA)

Abstract:
Somatic hybridization is an alternative in producing Dutch Elm Disease resistant elm but the introduction of selectable markers into the potential parents is needed. Genetic transformation should be used, provided that efficient plant regeneration system is available. We reported here, on in vitro plant regeneration from cambium of the resistant elm 'Sapporo gold 2'. Three to seven buds were induced per cambium explants, within 2–3 weeks and 1–3 shoots within 5–6 weeks. Shoot forming capacity extended from autumn to the end of spring. It was enhanced by auxins (0.5 μM) and inhibited by cytokinins but partially restored by charcoal suggesting a relation between seasonal budding capacity and hormonal content or cell sensitivity of the cambium. The use of DKW macronutrients (x¼) and agarose increased by two fold the number of buds / explant (6–7), reducing apex necrosis by 40%. However the number of shoots / explant remain to be improved.
