ISHS Acta Horticulturae 447: III International Symposium on In Vitro Culture and Horticultural  Breeding

IN VITRO MULTIPLICATION AND PROPAGATION OF A SLOVENIAN SELECTION OF SHALLOT (ALLIUM ASCALONICUM L. CV. POHORKA)

Authors:   J. Berljak, D. Benedicic
Abstract:
To improve and increase plant production of Slovenian selection of shallot (Allium ascalonicum L. cv. Pohorka), using a method for rapid shoot multiplication and bulb formation, effects of different types of growth regulators were investigated. Initial culture was established from basal stem plate explants on shoot induction BDS medium supplemented with 4.5 μM BA. Basal stem plates of in vitro produced shallot shoots were used for induction of the next shoot generation. Adding of 0.6 μM IAA increased average number of shoots per explant. Isolated shoots were induced to form bulbs on hormone-free BDS medium supplemented with activated charcoal and high concentration of sucrose (15%), or they were elongated on hormone-free half strength basal medium, and developed in plantlets with well formed roots.
