Abstract:
Protocols for micropropagation of Japanese persimmon have been proposed by many authors, but at present no reliable method have been defined for "Kaki Tipo" (PVNA), the most important cultivar in Italy, which appeared to be recalcitrant to in vitro tissue culture in previous tests.
In order to test culture establishment and first steps of shoot proliferation of this cultivar, axillary buds for each treatment obtained from dormant shoots of ‘Kaki Tipo’ has been cultured on (½N) MS medium gelified with Difco Bacto Agar (DBA)(7 g/l) and ICN-Gel Gro (Gellan gum)(GG)(2g/l), added with trans-Zeatin (t-ZEA) (5 and 10μM) and 6-Benzylamino purine (BA) (22 and 44μM). Results have been analysed separately in order to test the effect of gelifying agents, the two growth regulators, their concentrations and their interactions.
The best gelifying agent has been ICN-Gel Gro: after 50 days of culture noticeable differences regarded the percentage of shoots longer than 10mm (56.6% - average length 24.9±0.9 mm in GG; 3.8% - average 11.6±0.3 mm in DBA). Taking into account the interaction gelifying agent/growth regulator, the combination of GG with BA and then with t-ZEA gave the best results (68.4% and 44.7% of shoots longer than 10mm respectively), on the contrary explants on DBA added both with BA or t-ZEA did poorly, or at all, developed into long shoots.
In relation to growth regulator concentration, the best performances were linked to both levels of BA (22 and 44μM) (68.4% of shoots longer than 10mm), followed by 10μM and 5μM of t-ZEA when added to GG. On DBA only explants cultured with BA at 22μM developed into long shoots.
On GG with BA (22 and 44μM) proliferation rate on 30% of explants was 4±1.4 and 2.4±1.2 respectively.
No proliferation was noticed on Difco Bacto Agar, regardless the growth regulator added and when t-Zeatin was used.
