PROPAGATION OF POTATO USING TISSUE CULTURE TECHNIQUE

Abstract:
Using high concentration of auxin led to the callus production only and cytokinine did not play any role in this respect, cytokinine alone produced shoot formation without callus, low auxin concentration plus moderate concentration of cytokinine led to development of complete plantlets from meristem tips with avoid producing callus, high concentration of kn 2 mg/L and low concentration of NAA 0.1 mg/L produced callus plus 100% of shooting while high level of BA and low concentration of IBA induced callus formation without shoot proliferation.

Solid medium was better than the liquid medium for the meristem culture, plantlets raised in liquid medium had no root and 75% of meristems could produce shoot in liquid medium comparing with 100% in solid medium

Culturing nodal cutting in liquid medium reduced its efficiency as regarding percentage of root, shoot formation and shoot length. Data indicated that rooting may happen in nodal cutting of potato without using growth regulators, meanwhile the root characters generally were lower than those obtained when added hormones to the medium. BA or CCC plus BA led to the pushing of the potato plantlets to produce microtubers. CCC at 500 ppm plus BA at 5 ppm increased number and weight of microtubers as compared with using BA alone at 10 ppm
