PLANT REGENERATION AND SOMATIC EMBRYOGENESIS FROM SHOOT TIP AND COTYLEDON OF CUCUMIS MELO (GALIA)

Abstract:
Galia cantaloupe (Cucumis melo L.) explants including part of the cotyledon and shoot tip were used to produce somatic embryo and shoot organogensis. Two type of tissue were produced by cotyledon explant, a compact green organogenic tissue and green friable callus according to the supplemented with growth regulators. Adventitious shoots from the shoot tips were obtained in basal MS medium which contained BA at 2.0 mg/l. Kinetin was sufficient to development of shoots growth but did not affect on multiple axillary shoots. After 6 weeks incubation, micro cutting were successful rooted on media containing IBA or NAA at 2.0 mg/l for each and then transferred to a mixture of equal parts sand and peatmoss. Transparent plastic bags were inverted over the pots for one week to ensure high humidity around the newly transplanted plants.
